Gene Dosage and Galactose Utilization by Saccharomyces Tetraploids.

چکیده
In yeast, the enzymes required for steps (1) to ( 3 ) are “induced” enzymes which are synthesized only if galactose is present in the medium (DE ROBICHONSZULMAJSTER 1958). The nonallelic genes GA, and GA, are required in our strains of Saccharomyces for the rapid fermentation of galactose. GA, appears to control the transfer of galactose into the cell (DOUGLAS and CONDIE 1954) ; in the absence of GA,, utilization of this sugar is markedly dependent upon its concentration in the medium. The absence of GA, results, however, in a complete block of fermentation and respiration of galactose, since the cells cannot synthesize galactokinase (DE ROBICHON-SZULMAJSTER 1958). NELSON (1953) has shown that when glucose-grown cells are adapted aerobically to respire galactose, the required time of exposure to the inducer before rapid oxidation occurs decreases as the dosage of GA, increases. Further, in cells which are adapted anaerobically to galactose, the rate of fermentation also in-

Transformation and expression of Penicillium funicolusum glucose oxidase gene in yeast
Glucose oxidase is an important enzyme hydrolyzing for its hydrolyzing activity on glucos. It possesses and has a wide board of applications in different industries such as bakery, pharmaceutical, plant pathology and biosensors. In this study, yeast (Saccharomyces cerevisiae) was transformed successfully by the glucose oxidase gene (gox) obtained from Penicillium funicolusum. The secreted gluco...
متن کاملCLONING AND EXPRESSION OF HUMAN IFNα2B GENE IN SACCHAROMYCES CEREVISIAE
Interferon is a protein secreted by eucaryotic cells following stimulation by viruses, bacteria, and many other immunogenes. Recent medical studies indicate that interferons have effective role in the treatment of virus infections, immunodeficiency and certain types of cancer such as hairy cell leukaemia (HCL). The aim of the present study is to apply yeast strain for secreting human IFNα2b fol...
متن کاملGenetic co-regulation of galactose and melibiose utilization in Saccharomyces.
The gal3 mutation of Saccharomyces, which is associated with an impairment in the utilization of galactose, has been shown to be pleiotropic, causing similar impairments in the utilization of melibiose and maltose. Milibiose utilization and alpha-galactosidase production are directly controlled by the galactose regulatory elements i, c, and GAL4. The fermentation of maltose and the induction of...
متن کاملGalactose inhibition of the constitutive transport of hexoses in Saccharomyces cerevisiae.
The relationship between the pathways of glucose and galactose utilization in Saccharomyces cerevisiae has been studied. Galactose (which is transported and phosphorylated by inducible systems) is a strong inhibitor of the utilization of glucose, fructose and mannose (which have the same constitutive transport and phosphorylation systems). Conversely, all these three hexoses inhibit the utiliza...
متن کاملOngoing resolution of duplicate gene functions shapes the diversification of a metabolic network
The evolutionary mechanisms leading to duplicate gene retention are well understood, but the long-term impacts of paralog differentiation on the regulation of metabolism remain underappreciated. Here we experimentally dissect the functions of two pairs of ancient paralogs of the GALactose sugar utilization network in two yeast species. We show that the Saccharomyces uvarum network is more activ...
